RESUMO

In this study, a gene encoding for acetylxylan esterase was cloned and expressed in E. coli. A single uniform band with molecular weight of 31.2 kDa was observed in SDS-PAGE electrophoresis. Served as the substrate, p-nitrophenol butyrate was employed to detect the recombinant enzyme activity. It exhibited activity at a wide temperature range (30-100 °C) and pH (5.0-9.0) with the optimal temperature of 70 °C and pH 8.0. Acetylxylan esterase showed two substrates' specificities with the highest Vmax of 177.2 U/mg and Km of 20.98 mM against p-nitrophenol butyrate. Meanwhile, the Vmax of p-nitrophenol acetate was 137.0 U/mg and Km 12.16 mM. The acetic acid yield of 0.39 g/g was obtained (70 °C and pH 8.0) from wheat bran pretreated using amylase and papain. This study showed the highest yield up to date and developed a promising strategy for acetic acid production using wheat bran.

PURPOSE: Research on the concept of expressed emotion (EE) has expanded in recent years but its role in dementia still requires elucidation. Understanding the role of EE in the dementia context could help in the development of appropriate interventions. METHOD: The current review synthesized relevant literature to investigate the prevalence and correlates of EE status in families of people with dementia. A comprehensive search of four databases from inception to 2022 produced 2,683 papers; 18 studies met inclusion criteria. RESULTS: The use of EE criteria differed not only across cultural contexts, but even within the same cultural context. Overall, the prevalence of EE in families with dementia compared with other psychiatric conditions was not high. CONCLUSION: Specific changes in EE over time remain to be explored, and findings emphasize the need to carefully discriminate High EE status based on the cultural background of family members with dementia. The role of upflow velocity and Ca2+ concentration in controlling the type and rate of CaCO3 crystallization and their impacts on the anaerobic granular sludge (AnGS) formation and performance in an expanded granular sludge bed (EGSB) reactor were studied. The results showed that an improved upflow velocity could promote metastable CaCO3 crystals and achieve the optimized portion of vaterite with a value of 84 % at 10 m/h with a small amount of aragonite, thus limiting the scaling in the reactor. The removal efficiency of Ca2+ was to some extent positively correlated to the influent Ca2+ concentration, but declined when Ca2+ exceeded a specific threshold. Vaterite was dominant with the increase of Ca2+ concentrations of the influent. Compared with granules in R1 (Ca2+ 10 mg/L) and R2 (Ca2+ 100 mg/L), granules cultivated in R3 (Ca2+ 800 mg/L) revealed maximum amount of biomass with biggest particle size distribution and fastest average settling rate, with relative stable COD removal efficiency and the fast optimized reactor capacity at OLR of 16 kgCOD/m3d. A low upflow velocity and a higher Ca2+ concentration promoted nucleus formation and granules growth at the initial cultivation stage of the EGSB reactor. The Ca2+ concentration had a significant impact on the bacterial community and favoured the growth of Tolumonas and Anaeromousa Anaeroarcus. Archaea, rather than bacteria, was strengthened to contribute more to methane production at a relatively high Ca2+ concentration.

The rational design of absorber size is a promising strategy for obtaining excellent electromagnetic wave (EMW) absorption performance. However, achieving controllable tuning of the material size through simple methods is challenging and the associated EMW attenuation mechanisms are still unclear. In this study, the sizes of metal-organic frameworks (MOFs) are successfully tailored by changing the growth time and the molar ratio of iron (Fe)/organic ligands. The lateral and vertical lengths of MOFs vary in the range of 200 nm to 2 µm and 100 nm to 1 µm, respectively. Both experiments and simulations confirm that the decrease of MOF size favors the formation of more conductive networks, which is beneficial for improving the conductivity loss. Meanwhile, the micromagnetic simulation reveals that the magnetic coupling can be effectively enhanced by the decrease of MOF size, which is conducive to the improvement of magnetic loss, especially in low-frequency range. The reflection loss of Fe-based MOFs with optimized size reaches -46.4 dB at 6.2 GHz with an effective absorption bandwidth of 3.1 GHz. This work illustrates the important role of size effect in EMW dissipation and provides an effective strategy for enhancing the low-frequency EMW absorption performance.

Background: This study aimed to explore technetium-99m-diethylenetriaminepentaacetic acid (99mTc-DTPA) renal dynamicimaging to evaluate duplex kidney function in adult patients.Subjects and Methods: We retrospectively analyzed the clinical data of 25 patients with duplex kidneys who underwent 99mTc-DTPA renal dynamic imaging between June 2011 and March 2023 at our hospital. Patients in the duplex kidney group (n = 25)were divided into renogram normal (n = 9) and abnormal (n = 16) groups according to the imaging data. Additionally, normalpatients were selected as the control group (n = 25). After imaging, the region of interest of the kidneys was delineated, andrenography was performed. Renography can provide renal function parameters, including glomerular filtration rate (GFR),Tmax, T1/2, renal clearance, and the GFR ratio of the duplex renal segment (upper renal moiety).Results: Compared with the control group, the serum creatinine level in the duplex kidney group was higher (p = 0.025), GFRwas lower (p = 0.001), and patients with impaired renal function were mainly in the abnormal renography group (p = 0.001). Inthe duplex kidney group, the GFR (p = 0.026) and renal clearance (p = 0.006) of the affected kidneys were lower than those of thecontralateral kidneys, and Tmax (p = 0.025) was higher than that of the contralateral kidneys. There were no differences in renalfunction indicators of duplex renal segments with different GFR ratios. However, when the GFR ratio exceeded 50%, the renalfunction tended to decline.Conclusions: 99mTc-DTPA renal dynamic imaging was found useful to evaluate the total renal function, split renal function,and upper urinary tract patency in patients with duplex kidneys. Patients with abnormal renography results had worse renalfunction, and those with poor renal clearance in the affected renal moiety required surgical treatment. (AU)

High-value chemical production from natural lignocellulose transformation is a reliable waste utilization approach. A gene encoding cold-adapted carboxylesterase in Arthrobacter soli Em07 was identified. The gene was cloned and expressed in Escherichia coli to obtain a carboxylesterase enzyme with a molecular weight of 37.2 KDa. The activity of the enzyme was determined using α-naphthyl acetate as substrate. Results showed that the optimum enzyme activity of carboxylesterase was at 10 °C and pH 7.0. It was also found that the enzyme could degrade 20 mg enzymatic pretreated de-starched wheat bran (DSWB) to produce 235.8 µg of ferulic acid under the same conditions, which was 5.6 times more than the control. Compared to the chemical strategy, enzymatic pretreatment is advantageous because it is environmentally friendly, and the by-products can be easily treated. Therefore, this strategy provides an effective method for high-value utilization of biomass waste in agriculture and industry.

RESUMO

We reported a 23-year-old male patient with panhypopituitarism who underwent two resections for craniopharyngioma and received postoperative hormone replacement therapy. The 99mTc-MDP bone scan revealed focal high uptake of radioactive nuclide in multiple large joints. The SPECT/CT demonstrated the focal high uptake in their metaphysis. Thus, delayed epiphyseal closure was considered.

BACKGROUND: This study evaluated the preliminary effect of an integrated novel intervention comprising visualised sleep report feedback using information and communication technology and periodic health guidance on improving sleep indicators among community-dwelling older people. METHODS: The intervention was implemented among 29 older people in Sakai City, Japan, in a 3 months pilot trial. Non-worn actigraph devices were placed under participants' bedding to continuously measure their sleep state, and they received monthly sleep reports in writing. Sleep efficiency, total sleep time, sleep latency, and the number of times away from bed were recorded. A trained nurse expertly interpreted participants' sleep data and provided telephone health guidance. The first month's data were used as the baseline (T1), the second month provided data for the first intervention (T2), and the third month provided data for the second intervention (T3). Friedman tests and Wilcoxon signed-rank tests were used to examine differences in sleep outcomes between different time points. RESULTS: Participants' mean age was 78.97 ± 5.15 years, and 51.72% (15/29) were female. Comparison of T2 and T1 showed the intervention decreased participants' sleep latency at T2 (P = 0.038). Compared with T1, the intervention significantly decreased sleep latency (P = 0.004), increased total sleep time (P < 0.001), and improved sleep efficiency (P < 0.001) at T3. When T3 was compared with T2, only total sleep time was significantly increased (P < 0.001). There were no significant differences in the number of times away from bed across the three time points (P > 0.05). CONCLUSIONS: This visualised sleep report feedback and periodic health guidance intervention for community-dwelling older people showed promising, albeit small preliminary effects on sleep. A fully powered randomised controlled trial is required to verify the significance of this effect.

A 57-year-old woman was diagnosed as lung adenocarcinoma. 99mTc-MDP bone scan showed multiple radioactivity concentrated lesions on bilateral chest wall, which were confirmed as calcification foci after breast implant rupture by SPECT/CT. SPECT/CT could be used for differential diagnosis of breast implant rupture and malignant lesions.

OBJECTIVE: This review will determine the prevalence of sleep disturbances in individuals with mild cognitive impairment. INTRODUCTION: Mild cognitive impairment is regarded as a transitional state between normal functioning and dementia, and has a high likelihood of conversion to dementia. Individuals with mild cognitive impairment may suffer more severe sleep disturbances compared with normal older people. In some studies, sleep disturbances were associated with significantly higher odds of mild cognitive impairment. There is a need for prevalence estimates of sleep disturbances in people with mild cognitive impairment based on the currently available literature to guide clinical health care professionals and public health policies. INCLUSION CRITERIA: The review will consider studies reporting on the prevalence of sleep disturbances in individuals with mild cognitive impairment, using validated instruments, including subjective and/or objective measures. Studies will be excluded if the participants report sleep-related breathing or movement disorders. Studies using only the Mini-Mental State Examination to diagnose mild cognitive impairment will also be excluded. METHODS: The review will follow the JBI methodology for systematic reviews of prevalence and incidence. The MEDLINE (Ovid), Embase, Cochrane Library (CDSR and CENTRAL), CINAHL (EBSCOhost), PsycINFO (EBSCOhost), Scopus, and Web of Science Core Collection databases will be systematically searched from inception to the present with no language limitations. Analytical observational studies, including prospective and retrospective cohort, case-control, and cross-sectional studies, will be considered. Two reviewers will independently conduct the study selection, critical appraisal, and data extraction. Methodological quality will be evaluated using the JBI critical appraisal checklist for studies reporting prevalence data. A meta-analysis will be conducted to synthesize the prevalence data, where possible. REVIEW REGISTRATION: PROSPERO CRD42022366108.

The aim of this work is to study a bifunctional endoglucanase/carboxylesterase in Sphingobacterium soilsilvae Em02 and express it in soluble form in engineered Escherichia coli. The molecular weight of the recombinant protein of the bifunctional enzyme was 41 KDa. This research also determined the enzymatic activities of the bifunctional enzymes using microcrystalline cellulose and p-nitrophenyl butyrate as substrates and found 40 °C as the optimum temperature for their enzymatic activities. The optimal pH in dual function was 6.0 for endoglucanase and 7.0 for carboxylesterase. The bifunctional enzyme also exhibited enzymatic activities on the natural biomass by generating up to 3.94 mg of glucose and 49.4 µg of ferulic acid from 20 mg of destarched wheat bran. This indicates the broad application prospects of the bifunctional enzyme in agriculture and industry.

Anammox granule sludge (AnGS) has great potential in the field of wastewater nitrogen removal, but its development and promotion have been limited by the slow granulation speed and fragile operating stability. Based on the reviews about the AnGS formation mechanism in this paper, "micro-nuclei" was found to play an important role in the granulation of AnGS, and adding "micro-nuclei" directly into the reactor may be an efficient way to accelerate the formation of AnGS. Then, accelerating AnGS granulation with inert particles, multivalent positive ions, and broken granule sludge as "micro-nuclei" was summarized and discussed. Among inert particles, iron-based particles may be a more advantageous candidate for "micro-nuclei" due to their ability to provide attachment sites and release ferric/ferrous ions. The precipitations of multivalent positive ions are also a potential option for "micro-nuclei" that can be generated in-situ, but a suitable dosing strategy is necessary. About broken granular sludge, the broken active AnGS may have advantages in terms of anaerobic ammonium oxidation bacteria-affinity and granulation speed, while using inactive granular sludge as "micro-nuclei" can avoid interfering bacterial invasion and has a higher cost performance than broken active AnGS. In addition, possible research directions for accelerating the formation of AnGS by dosing "micro-nuclei" were highlighted. This paper is intended to provide a possible pathway for the rapid start-up of AnGS systems, and references for the optimization and promotion of the AnGS process.

RESUMO

We report a case of accidental local 99mTc-MDP uptake in left lung on bone scan without pulmonary parenchymal abnormality on SPECT/CT. The abnormal uptake disappeared on repeated bone scan and SPECT/CT after 3 days. It is speculated that the high radioactivity focus is a microthrombus in pulmonary artery.

It has been accepted that the uniform distribution of magnetic metal particles is beneficial to microwave absorption, while why the homogeneous magnetic particles on the dielectric substrate improve the electromagnetic loss is still unclear. Herein, metal Ni nanoparticles, two-dimensional (2D) basalt/scattered Ni, and basalt/uniform Ni microflakes are obtained through a pretreatment and electroless deposition process. In comparison to Ni nanoparticles and basalt/scattered Ni, the basalt/Ni microflakes with largely uniform and compact Ni nanolayers on basalt, breaking the percolation limit, are favorable for enhanced electromagnetic attenuation. The Ni nanolayers are convenient for construction of a microscale conductive net and migration of an electron. The 2D heterostructures constructed by basalt substrates and decorated Ni layers boost multiple scattering absorption and promote interfacial polarization. Meanwhile, exposed Ni does not inhibit magnetic resonance, enabling strong magnetic coupling. Consequently, the basalt/Ni microflakes with uniform Ni nanolayers demonstrate better microwave absorption with a minimum reflection loss of -30 dB and an effective absorption bandwidth of 3 GHz at 1 mm. This work shows that the uniform and compact magnetic metal nanolayers are effective in improving the dielectric loss and magnetic loss simultaneously to achieve the high-performance microwave absorption.

Developing megahertz (MHz) electromagnetic wave (EMW) absorption materials with broadband absorption, multi-temperature adaptability, and facile preparation method remains a challenge. Herein, nanocrystalline FeCoNiCr0.4Cu0.2 high-entropy alloy powders (HEAs) with both large aspect ratios and thin intergranular amorphous layers are constructed by a multistage mechanical alloying strategy, aiming to achieve excellent and temperature-stable permeability and EMW absorption. A single-phase face-centered cubic structure with good ductility and high crystallinity is obtained as wet milling precursors, via precisely controlling dry milling time. Then, HEAs are flattened to improve aspect ratios by synergistically regulating wet milling time. FeCoNiCr0.4Cu0.2 HEAs with dry milling 20 h and wet milling 5 h (D20) exhibit higher and more stable permeability because of larger aspect ratios and thinner intergranular amorphous layers. The maximum reflection loss (RL) of D20/SiO2 composites is greater than - 7 dB with 5 mm thickness, and EMW absorption bandwidth (RL < - 7 dB) can maintain between 523 and 600 MHz from - 50 to 150 °C. Furthermore, relying on the "cocktail effect" of HEAs, D20 sample also exhibits excellent corrosion resistance and high Curie temperature. This work provides a facile and tunable strategy to design MHz electromagnetic absorbers with temperature stability, broadband, and resistance to harsh environments.

Electromagnetic (EM) absorbers serving in the megahertz (MHz) band and a wide temperature range (from -50 to 150 °C) require high and temperature-stable permeability for outstanding EM absorption performance. Herein, FeCoNiCr0.4CuX high-entropy alloy (HEA) powders with a unique nanocrystalline structure separated by a thin amorphous layer (NTA) are designed to improve permeability and enhance intergranular coupling. Simultaneously, the long-range anisotropy is introduced via devising the preparation process and tuning the chemical composition, such that the intergranular exchange interaction is further strengthened for stable permeability and EM wave absorption in a wide temperature range. FeCoNiCr0.4Cu0.2 HEAs exhibit a near-zero permeability temperature coefficient (5.7 × 10-7 °C-1) a in wide temperature range. The maximum reflection loss (RL) of FeCoNiCr0.4Cu0.2 HEAs is higher than -7 dB with 5 mm thickness at -50-150 °C, and the absorption bandwidth (RL < -7 dB) can almost cover 400-1000 MHz. Furthermore, FeCoNiCr0.4Cu0.2 HEAs also have a high Curie temperature (770 °C) and distinguished oxidation resistance. The permeability temperature dependence of FeCoNiCr0.4CuX HEAs is investigated in-depth in light of the microstructural change induced by tuning the chemical composition, and a new inspiration is provided for the design of magnetic applications serving in wide temperature, such as transformers, sensors, and EM absorbers.

Diet and lifestyle choices contribute to obesity and liver disease. Broccoli, a brassica vegetable, may mitigate negative effects of both diet and lifestyle. Currently, there are no clinically relevant, established molecular biomarkers that reflect variability in human absorption of brassica bioactives, which may be the cause of variability/inconsistencies in health benefits in the human population. Here, we focused on the plasma metabolite profile and composition of the gut microbiome in rats, a relatively homogenous population in terms of gut microbiota, genetics, sex and diet, to determine if changes in the plasma metabolite profiles caused by dietary broccoli relate to molecular changes in liver. Our aim was to identify plasma indicators that reflect how liver health is impacted by dietary broccoli. Rats were fed a 10% broccoli diet for 14 days. We examined the plasma metabolite composition by metabolomics analysis using GC-MS and gut microbiota using 16S sequencing after 0, 1, 2, 4, 7, 14 days of broccoli feeding. We identified 25 plasma metabolites that changed with broccoli consumption, including metabolites associated with hepatic glutathione synthesis, and with de novo fatty acid synthesis. Glutamine, stearic acid, and S-methyl-L-cysteine (SMC) relative abundance changes correlated with changes in gut bacteria previously implicated in metabolic disease and with validated increases in expression of hepatic NAD(P)H dehydrogenase [quinone] 1 (NQO1) and nuclear factor (erythroid-derived 2)-like 2 (Nrf2), associated with elevated hepatic glutathione synthesis. Circulating biomarkers following broccoli consumption reflect gut-liver axis health.

Geographic food fraud - misrepresenting the geographic origin of a food item, is very difficult to detect, and therefore this type of fraud tends to go undetected. This potentially negatively impacts the health of Canadians and economic success of our seafood industry. Surveillance studies have shown that up to a significant portion of commercially sold seafood items in Canada are mislabeled or otherwise misrepresented in some way. The current study aimed to determine if the microbiome of fresh shellfish could be used as an accurate marker of harvest location. Total DNA was extracted from the homogenate of 25 batches of fresh soft-shell clams (Mya arenaria) harvested in 2015 and 2018 from two locations on the East Coast of Canada and the microbiome of each homogenate was characterized using 16S rRNA targeted amplicon sequencing. Clams harvested from Nova Scotia in both years had a higher abundance of Proteobacteria and Acidobacteria (p < 0.05), but a lower abundance of Actinobacteria (p < 0.05) than those from Quebec. Alpha-diversity also differed significantly between sites. Samples harvested from Nova Scotia had greater diversity (p < 0.0001) than those from Quebec. Beta-diversity analysis showed that the microbial community composition was significantly different between the samples from Nova Scotia and Quebec and indicated that 16S rRNA targeted amplicon sequencing might be an effective tool for elucidating the geographic origin of unprocessed shellfish. To evaluate if the microbiome of shellfish experiences a loss of microbial diversity during processing and storage - which would limit the ability of this technique to link retail samples to geographic origin, 10 batches of retail clams purchased from grocery stores were also examined. Microbial diversity and species richness was significantly lower in retail clams, and heavily dominated by Proteobacteria, a typical spoilage organism for fresh seafood, this may make determining the geographic origin of seafood items more difficult in retail clams than in freshly harvested clams.

As a phosphorus-rich material, low-temperature combustion sewage sludge ash (LTCA) contains over 9 wt% content of phosphorus (P) and a large proportion of impurities, especially the content of Fe arrives at 14.59 wt%. To fully utilize LTCA as a fertilizer, this study investigated a procedure for P recovery from LTCA via struvite crystallization with fewer impurities. The adsorption characteristics of P and Fe by cation exchange resin (CER) were explored by simulating using the macroscopic parametric equation Thomas model. Optimum purification conditions for P-rich leachate by cation exchange column method were determined. Results showed that approximately 97.21 wt% of P was extracted from LTCA at HCl concentration of 0.8 M and liquid/solid ratio of 20.0 ml/g. More than 90 wt% of impurities could be detached by making P-rich leachate flow through cation exchange bed filled by CER at 300 ml/h. The macroscopic parametric equation Thomas model could clearly describe the adsorption characteristics of Fe in P-rich leachate by CER. Theoretical basis for purification of high concentration Fe in P-rich leachate by CER was provided. Approximately 84.04 wt% of total P in LTCA was recovered as struvite crystal which had low concentrations of heavy metals (5.96 mg/kg for Cr, 45.21 mg/kg for Cu, 29.67 mg/kg for Ni, 2.24 mg/kg for Pb, and 290.6 mg/kg for Zn) and could be eco-friendly for agricultural application. X-ray diffraction and SEM-EDS analysis validated the formation of struvite.
